Response to any ITP treatment is so individual, it's not wise to think in terms of what "usually" happens.
All I can say is that on the day of my first NPlate dose, my count was 15. Subsequent weeks were 60, 345, 115, 1, 100, 249, 210, 3... !
This has been my wild and crazy response to NPlate for three years now. At least now, the highs are lower and the lows are around the 50 target. At the time I started it, I was also taking prednisone, around 15-20 mg a day, which probably caused a boost in effectiveness as well.

Each injection takes 4 or 5 days to take effect so no point in doing more frequent bloods than weekly. Apart from that it's very individual. It took me 6 months to get stable on it and about 3 years to be able to come off in a sort of remission. Relapses have only needed a few months to get back into partial remission.
