I am having extensive dental work done tomorrow so had a gamma transfusion last Friday. I had the blood test yesterday where for some reason they wanted two tubes. They were taken literally seconds after one another and each tube had a different count - 242 and 250. This is not a concern but just shows plt counts can vary in that short while.

Yep - the number you get is just a sample....a close estimate. Sometimes other cells that happen to be a similar size to platelets sneak in there and skew the number. I'm glad your counts are up!
