Lman, I think I know what you are saying. Taking 375 every 5 or 6 days so that the average dose over 7 days would be 500? Then there is nothing to throw away. I don't recall anyone here even considering that sort thing. Anne, Posey, anyone else have thoughts on this?
Are the vials exactly 375? Or, do they just have a minimum of 250mcg ?

I've copied and pasted this from UK 'electronic medicines compendium' website because I'm not sure people outside the UK can access it.
:-Nplate 250 micrograms powder and solvent for solution for injection
Each vial contains 250 mcg of romiplostim. After reconstitution, a deliverable volume of 0.5 mL solution contains 250 mcg of romiplostim (500 mcg/mL). An additional overfill is included in each vial to ensure that 250 mcg of romiplostim can be delivered.

Yes, it does feel wasteful, but once reconstituted is only good for 24 hrs. Once a vial of Nplate is used, that's it. The rest must be thrown out.

It has also written in the leaflet inside each Nplate box that each 250 mcg contains 375 mcg and each 500 mcg contains 625 mcg indeed.
I have also noticed that if you solve 0.72 mcg in a 250 mcg vial, you get more than 0.5 ml which leaflet says you must inject it, so this also says there's more than 250 mcg of romiplostim in each 250 mcg vial.

Ok, looking at table one in the link, I see what you are saying. Both 250 and 500 vials have an extra 125mcg in them.
Still thinking about taking one vial, with 'overfill', every 5 or 6 days to get a higher average dose over 7 days?

I don't know Hal, I don't want to be back to the hospital and I'm cautious.
Yesterday I was 260k. I think I used too much of romiplostim.
I mixed two vials and injected 2/3 of it to myself, so that's (if we say each vial has 375mcg) : 2*375*(2/3)=500 mcg
and I'm 80kg, so it's 500/80=6.25
I hope I can return to my previous regimen. I'll follow Amgen's dosing guideline.

Lman, all Romiplostim vials have overfill to make sure the 250 or 500 doses can be administered.

Hello every one.
I have switched to once a month tests, I currently use 500mcg each week :(
my previous counts: 130k,98k,110k.
By the way, I had a question. Couple of days after I first increased my dose, I have had severe itching, which was considerably lowered after a couple of weeks. But now I still "feel" I sometimes itch. I have had no other skin symptoms or any other symptoms.
I have read that pruritus skin might be as common as 10% in nplate users, but I have not had this problem while I was on 375mcg.
And to be honest, whenever I think to itch, I itch! But when I'm doing my job and don't pay attention to it, I don't. I told my condition to the doctor and he told it's ok and it's probably an allergic reaction and did nothing. What shall I do?
How much is itching is normal for a human? because I'm not sure wheater this amount of itching is abnormal or not.
Has anyone had itching problem with nplate?

Is it possible this itching actually became a problem when counts crashed and not when Nplate dose was increased?
