Has anyone taken Danazol in combination with NPlate?
I am wondering how well it worked.

Meredith, I am taking both Danazol and Promacta.

As you probably know Danazol reduces platelet destruction and Promacta increases platelet production. But when one combines the two, there is a multiplying affect. That is, instead of being an additive affect on counts, it is more synergistic. It works a bit better than either alone.

Let me give a hypothetical numeric example. If a certain dose of Danazol alone raised counts by 40 (from baseline), and, a certain dose of Promacta raised counts by 40 (from baseline), taking both will not raise counts by 80. It will be more like 80 + 20% = 96.

For me. I am taking 100 mg of Danazol and half of a 12.5 mg tablet of Promacta per day. This combination gives a typical count just above 50. Thinking about it with the above example in mind. A full 12.5 mg tablet alone gives me about the same count. But 200 mg of Danazol gave me a 37 count. So an additive effect would have been about 10 (baseline) + 20 (promacta increase) + 14 (danazol increase) = 44. But the count is actually more like: 44 + 20% = 53.

Does that help? LOL, perhaps I'm completely off base and you were actually asking about symptoms?

Thank you. I take Danazol with NPlate. So I guess we are talking about the same theory. The numbers still jump all around. Promacta’s copay would be $2500. Per month. Do you know of any way to get around that? I applied for financail aid for another drug and was denied.

Meredith, hope the combination works for you. Yes, same theory.

If you haven't looked up Danazol before, be aware that about 20% of folks have problems with liver enzymes (AST and ALT) going out of range high with it. This happened to me. A dose of 100 mg is the highest I can take.

When trying steroids, if you had a weaker steroid response, be sure to give Nplate max dose (= 10) lots of time to work. For some folks, they end up responding and need less and less of a dose over time/months. Sometimes a bit of steroids with Nplate can speed the process.
